Biopharmaceutical Aspects

From molecular properties to clinical performance — how particle size shapes the fate of a drug in the body.

Biopharmaceutical Aspects

Micronization serves as a fundamental particle engineering technique, typically reducing the average particle size of APIs to less than 10 microns. In the journey of drug development, it represents the critical juncture where Drug Substance meets Drug Product, transforming raw crystalline materials into engineered particles with optimized clinical performance. The primary biopharmaceutical driver for this process is the enhancement of bioavailability for poorly soluble molecules, often categorized as BCS Class II compounds. According to the principles of the Noyes-Whitney equation, the dissolution rate of a drug is directly proportional to its available surface area. By decreasing the geometric particle size, micronization exponentially increases the specific surface area, significantly increasing the dissolution rate for orally administered drugs. This acceleration in dissolution kinetics facilitates more rapid absorption, resulting in higher plasma peak concentrations (Cmax) and improved overall exposure as measured by the area under the curve (AUC).

These improvements directly contribute to higher therapeutic efficiency, often enabling dose reduction without compromising clinical outcomes. This is particularly advantageous for minimizing dose-dependent side effects. Furthermore, micronization is vital for specialized delivery routes beyond oral administration. In pulmonary delivery, it is utilized to improve the penetration capacity of solid particles in the airways by targeting a precise respirable aerodynamic diameter, generally between 1 and 5 μm. This precision ensures that the drug reaches the peripheral airways where absorption is most effective.

For ophthalmic applications, reducing particle size is a key strategy to improve product tolerability, ensuring that the presence of coarse particles does not cause mechanical irritation to sensitive ocular tissues (e.g. sand-in-the-eye feeling).

The technique also provides significant advantages for topical formulations by increasing the covering capacity of creams and ointments, allowing for a more effective and uniform distribution of the drug on the skin surface.

By ensuring material homogeneity and the achievement of target physical specifications, micronization ensures consistent biopharmaceutical performance across a wide range of therapeutic areas.
